Anti-Risankizumab Antibody (3A11) inhibits the binding of Risankizumab to IL-23.

Human IL23A Protein was coated at a concentration of 1 μg/mL, followed by the addition of Risankizumab at 0.6 μg/mL and Anti-Risankizumab antibody that was serially diluted starting from 10 μg/mL. Detection was performed using Mouse Anti-Human IgG Fc Antibody (50B4A9)[HRP], mAb (GenScript, A01854) diluted at 1:10,000.
MonoRab™ Anti-Risankizumab Antibody (3A11), mAb, Rabbit (GenScript, A02321) demonstrated inhibitory properties with an IC50 value of 0.69 µg/mL.

In this Sandwich ELISA assay, MonoRab™ Anti-Risankizumab Antibody (3A11), mAb, Rabbit (GenScript, A02321) was coated at a concentration of 1 μg/ml, and Mouse Anti-Human IgG Fc Antibody (50B4A9)[HRP], mAb (GenScript, A01854) was used as a detection antibody at a dilution of 1:10,000.
In this assay, a four-parameter logistic curve fitting program was used to create a standard curve with the R-Square equal to 0.99999. The typicaldynamic range of the assay is 0.14 - 11 ng/mL.

The MRD is the minimum dilution necessary for the detection of Risankizumab in biological matrix with least interference. Serum samples from cynomolgus monkey were serially diluted to determine the MRD of this assay, and the test result suggested that MRD was as 1:10.
In this ELISA assay, MonoRab™ Anti-Risankizumab Antibody (3A11), mAb, Rabbit (GenScript, A02321) was coated at a concentration of 1 μg/ml, and MonoRab™ Anti-Risankizumab Antibody (32A12), mAb, Rabbit (GenScript, A02322) conjugated with Biotin was used as a detection antibody at a concentration of 0.5 μg/mL.

Specificity analysis of the detection of Risankizumab.

Risankizumab standard samples at 6 concentrations were spiked with 10 µg/mL of human IgG1. The test result demonstrated that the high concentration of human IgG1 did not interfere with the detection of Risankizumab.

Storage Instructions The lyophilized product remains stable for up to 1 year at -20 °C from the date of receipt. Upon reconstitution, it can be stored for 2-3 weeks at 2-8 °C or for up to 12 months at -20 °C or below. Avoid repeated freezing and thawing cycles.

Target Background Risankizumab is a monoclonal antibody designed to target interleukin-23 (IL-23), a key cytokine involved in the inflammatory process of certain autoimmune diseases. By specifically binding to the p19 subunit of IL-23, Risankizumab inhibits its interaction with the IL-23 receptor, thereby reducing inflammation and immune responses. This mechanism makes Risankizumab particularly effective in the treatment of moderate to severe plaque psoriasis. The drug is marketed under the brand name Skyrizi and has shown superior efficacy compared to other biologics in its class. It is also being studied for potential use in other inflammatory conditions, such as Crohn's disease.
