Background Human CD32 group consists of FcγRIIa, b and c proteins that share 94-99% amino acid identity in their extracellular domains but differ substantially in their transmembrane and cytoplasmic domains. CD32B, also known as FcγRIIb, is a low affinity receptor that recognizes immunoglobulin gamma complexes. It is encoded by the FCGR2B gene and expressed on B cells and most myeloid cells. CD32B has an intrinsic cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM) and delivers an inhibitory signal upon ligand binding. Co-ligation of CD32B with B cells antigen receptor (BCR) results in selective dephosphorylation of CD19 tyrosine, tyrosine phosphorylation of the ITIM in the CD32B cytoplasmic domain, and recruitment of SH2-containing inositol 5-phosphatase (SHIP). This causes a block in calcium influx, inhibition of B cell activation, and leads to apoptosis. CD32B is a potential target for monoclonal antibody therapy for malignancies. Targeting CD32B may represent a novel approach to immunotherapy of B-cell malignancies.
