CHO-K1/BTLA Stable Cell Line

Figure 1. FACS analysis of cell surface expression of human BTLA on CHO-K1/BTLA cells. The CHO-K1/BTLA cells (blue) and the negative control CHO-K1 cells (red) were probed using goat anti-human BTLA antibody (Cat. No. AF3354, R&D Systems) and FITC-conjugated donkey anti-goat IgG (Cat. No. sc-2024, Santa Cruz).

CHO-K1/BTLA Stable Cell Line

The B and T lymphocyte attenuator (BTLA) is an Ig super family protein with an intermediate type Ig fold in the ectodomain and an ITIM inhibitory signaling domain in the cytosol. BTLA interacts with the herpesvirus entry mediator (HVEM; TNFRSF14), a TNFR super family member. Engagement of BTLA by HVEM, induces tyrosine phosphorylation of the ITIM motifs in the cytoplasmic tail of BTLA, allowing the recruitment of the phosphatases SHP-1 and SHP-2, which attenuate signaling.

Product Description Recombinant CHO-K1 cells stably overexpress human B and T lymphocyte associated (BTLA) on the surface. The surface expression of BTLA is validated by FACS analysis. This cell line is recommended for cell-based binding assay to screen antibodies against BTLA or to measure binding affinity between BTLA and anti-BTLA antibodies.
Culture Properties Adherent.
Stability Stability: Stable through more than 16 passages without significant changes in assay performance or expression profile.
Size Two vials of frozen cells (>1×106 per vial in 1 mL).
Storage Store cells in liquid nitrogen immediately upon receipt. Thaw and recover cells within one year from the date received.

Culture Medium Ham’s F-12K (Kaighn’s), 10% FBS, 600 μg/ml Geneticin (Cat. No. 10131-035, Life Technologies).
Complete Growth Medium Ham’s F-12K (Kaighn’s) (Cat. No. 21127-022, Life Technologies), 10% FBS (Cat. No. 10099-141, Life Technologies).
Freeze Medium-DATA 95% complete growth medium, 5% (V/V) DMSO (Cat. No. D2650, Sigma).
