Figure 1. Vasoactive intestinal peptide (VIP)-induced stimulation of intracellular calcium mobilization in HEK293/Gα15/VPAC2 cell pool. The cell pool was generated by transient transfection of VPAC2 plasmid on HEK293/Gα15 cells. Then cell pool were loaded with Calcium-4 prior to being stimulated with VAPC2 receptor’s agonist VIP. The intracellular calcium change was measured by FLIPR (Molecular Devices).

HEK293/Gα15 Stable Cell Line

HEK293/Gα15 is a HEK293 cell line stably expressing the Gα15 alpha subunit protein which a Gq protein.  It is used as a host cell for transfection expression of Gs and Gi/o -coupled receptors, the constitutively expressed Gα15 protein in the cells allows many transfected receptors which normally stimulate/inhibit the cAMP pathway, to couple to Gq signal transduction and mobilize intracellular calcium.  The cell line carries the puromycin resistance gene and is resistant to puromycin

Product Description Recombinant HEK293 cells stably overexpress human G protein subunit alpha 15 (Gα15) in the cytoplasm. This protein enables G protein - coupled receptor binding activity and acts upstream of or within with a positive effect on calcium - mediated signaling.
Culture Properties Adherent.
Stability Stable through more than 16 passages with no significant changes in assay performance or expression profile.
Size Two vials of frozen cells (>1×106 per vial in 1 mL).
Storage Store cells in liquid nitrogen immediately upon receipt. Thaw and recover cells within one year from the date received.

Culture Medium DMEM, 10% FBS, 0.5 μg/ml Puromycin B (Cat. No. A11138, Life Technologies)
Complete Growth Medium DMEM (Cat. No. 10566, Life Technologies), 10% FBS
Freeze Medium-DATA 90% FBS (Cat. No. 10099-141, Life Technologies), 10% DMSO (Cat. No. D2650, Sigma)
