Regeneration performance of MonoRab™ Anti-VHH Affinity Magnetic Beads
The target VHH was captured by GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads (Cat. No. L01034). The 100 μL settled beads were washed twice with 1 mL TBS and then eluted with 1 mL acidic eluent (0.1 M glycine HCl, pH 2.5). Subsequently, the beads were regenerated by 1 mL regeneration buffer for reuse. The coefficient of variation (CV) of the amount of eluted target protein was less than 20% for a total of 10 replicate purifications indicating that GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads is capable of multiple repeated use with relatively high consistency when eluted by acidic eluent.
Purification of humanized VHH from E.coli whole cell lysate
The humanized VHH (M.W.=15 kDa) from E.coli whole cell lysate was captured by GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads (Cat. No. L01034). The captured humanized VHH was subsequently eluted using an acidic eluent (0.1 M glycine HCl, pH 2.5) and subjected to SDS-PAGE analysis. The results showed that GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads are suitable for high-specificity purification of humanized VHH from E.coli whole cell lysate

Purification of camelid VHH from various lysates
The camelid VHH (M.W.=15 kDa ) from E.coli whole cell lysate, yeast whole cell lysate or mammalian cell culture supernatant was captured by GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads (Cat. No. L01034). The captured camelid VHH was subsequently eluted using an acidic eluent (0.1 M glycine HCl, pH 2.5) and subjected to SDS-PAGE analysis. The results showed that GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads are suitable for high-specificity purification of camelid VHH from widely used protein expression systems.
